Renin: kidney-derived enzyme that controls blood pressure
Renin is an enzyme produced by the kidneys that initiates the renin–angiotensin–aldosterone system, regulating blood pressure, fluid balance and electrolyte homeostasis.
Overview
Renin is a proteolytic enzyme released into the bloodstream by specialized kidney cells. It begins a hormone cascade called the renin–angiotensin–aldosterone system (RAAS) that helps adjust blood pressure, circulating volume and salt balance. More simply, renin converts a plasma protein into a peptide that ultimately narrows blood vessels and stimulates salt retention.

Renin is produced and secreted primarily by juxtaglomerular cells in the kidneys. As an enzyme, it cleaves the liver-derived substrate angiotensinogen to form angiotensin I. Subsequent steps in the circulation generate angiotensin II, a potent effector that raises pressure and influences renal sodium handling.
Regulation
Renin release is tightly regulated. Major stimuli include reduced renal perfusion pressure, decreased sodium delivery to the distal tubule, and sympathetic nervous system activation. Conversely, high blood pressure, increased sodium and some hormonal signals suppress renin secretion.
Clinical importance and therapies
Because renin is a key initiator of RAAS, it is central to understanding hypertension, heart failure and some kidney diseases. Several drug classes affect this system: direct renin inhibitors, angiotensin-converting enzyme inhibitors and angiotensin receptor blockers each intervene at different steps. Measuring renin activity or concentration can sometimes assist diagnostic evaluation of blood pressure disorders.
