Tuolumne County is a county located in the Sierra Nevada region of the U.S. state of California. Established in 1850, it has a long association with the California Gold Rush era and retains a largely rural character. The county seat and only incorporated city is Sonora. At the 2010 census the population was 54,179.

The county occupies foothills and mountain terrain rising into the high Sierra. It includes river valleys, conifer forests and granite ridgelines.
