How it works and characteristics

Propofol produces rapid loss of consciousness by enhancing inhibitory signalling in the central nervous system, primarily at GABA-A receptors. Its onset is fast — typically within about 30–60 seconds after an intravenous bolus — and recovery is relatively quick once administration stops, which makes it valuable for short procedures and for titratable sedation.

Administration and monitoring

Propofol is given only by trained clinicians via intravenous injection, either as single boluses or as a continuous infusion. Because it depresses breathing and lowers blood pressure, patients require airway support, oxygen and continuous cardiovascular and respiratory monitoring during use. Dosing is individualized and weight-based.

It interacts with other central nervous system depressants and should not be used outside monitored medical environments. Allergy to components of the emulsion (such as egg or soybean derivatives) is considered when choosing alternatives.

History and regulation

Developed in the late 20th century, propofol became popular because of its predictable recovery profile. Its potency and rapid action mean its distribution and use are tightly controlled and limited to clinical settings staffed by professionals trained in airway and cardiopulmonary management.
